Ubiquitination

Ubiquitination: labelling the proteins

In the late 70’s and early 80’s, ground-breaking research led to the discovery of a highly-specific, energy-dependent process enabling the cell to target specific proteins for degradation. In 2004, Aaron Ciechanover, Avram Hershko and Irwin Rose were awarded the Nobel Prize in Chemistry for their discovery, which enabled the understanding of protein degradation at the molecular level as well as its implication in biological processes as diverse as control of the cell cycle, gene transcription and immunity (1, 2

UPDATE: Apple Captures 95% Of Tablet Market In 3Q - WSJ.com

By Ian Sherr Of DOW JONES NEWSWIRES

SAN FRANCISCO (Dow Jones)--Apple Inc. (AAPL) dominated the global market for tablets in the third quarter amid limited competition for its iPad device, a situation that is likely to change, especially as netbook interest wanes.

Roughly 95% of all tablet computer shipments in the quarter were of the Cupertino, Calif. consumer electronics giant's tablet, according to a new study by Strategy Analytics Inc. The Boston-based research firm said the tablet computer shipments rose 26% from the previous quarter.

Google Inc.'s (GOOG) Android operating system represented just 2% of global tablet shipments that were made by generic device manufacturers. However, that percentage is expected to rise in the current quarter, as more models, such as Samsung Electronics Co.'s (005930.SE, SSNHY) Galaxy Tab, enter the market, the study said.

Apple has been selling its iPad touchscreen device since April with little competition from major competitors. Hewlett-Packard Co. (HPQ) has announced its plans to release tablets in the near future, along with display-maker Viewsonic Corp. Others, such as Research in Motion Ltd. (RIMM), have announced plans to release tablet devices sometime next year.

Election Day 2010: The Great Refudiation of Obama

Election Day 2010: The Great Refudiation of Obama

David Karki

Refute:  To prove wrong by argument or evidence, to show to be false or erroneous.

Repudiate:  To refuse to have anything to do with, to disown, to refuse to accept, to reject as unauthorized, untrue, unjust, or having no binding force

Reeeeee-jected.

Refudiate:  A new term coined by Alaska Governor Sarah Palin, which combines the above two words. And what will be done by voters to President Obama on Election Day, so as to demonstrate their anger at his, Speaker Pelosi’s, and Harry Reid’s having forced down America’s throat destructive and wrong liberal policies that have wrecked the economy and threaten our future well-being as a nation.

Don’t be fooled by the propagandist spin of a blatantly partisan liberal media that will defend and prop up at all costs the phony “messiah” they created back in 2008. This is a massive election defeat for the Democrats, of unprecedented historical proportions. Going from near veto-proof super-majorities to losing the House by a comfortable margin and very possibly the Senate as well, in a year when most of the seats up were on friendlier and safer liberal blue turf, is a staggering punch in the gut. To use poker lingo, they badly overplayed a hand that was much weaker than they thought, and now they’re about to lose most of their chips for it.

Don't forget to vote!

modern architecture — Infoplease.com

modern architecture

modern architecture, new architectural style that emerged in many Western countries in the decade after World War I. It was based on the “rational” use of modern materials, the principles of functionalist planning, and the rejection of historical precedent and ornament. This style has been generally designated as modern, although the labels International style, Neue Sachlichkeit, and functionalism have also been used.

Wood Shop Tip of the Day

Shop Tip of the Day

Hinge swings into action when holes require spacing
Drilling a series of accurately spaced holes can be grueling. Even a small misalignment becomes a glaring error in a row.Line up those holes smartly with a simple spacing jig for your drill press. Attach a fence (thicker than the material you'll be drilling) to the wooden auxiliary table of your drill press. Drill out the end hinge hole on the long arm of a T-hinge to accept a machine screw the diameter of the hole you'll be drilling. Now, secure the hinge to the fence top so the distance from the fence to the screw equals the distance from the edge of the board to the hole location. Clamp the jig to the drill-press table so the distance from the bit to the screw is the same as the space between the holes, and the distance from the bit to the fence equals the distance from the edge of the board to the hole. With the jig in place, measure, mark, and drill the first hole. Now, slide the workpiece along the fence. The screw will fall into the hole to stop the board in position for the next hole. Repeat to drill the rest of the holes.--Ed Abrams, Greenville, N.Y.

Thickness Planer – Reviews, Blades, Snipe, Dust Collection

A thickness planer allows you to plane the surface of a piece of wood to smooth out a poorly finished surface, mill the piece to a custom thickness, and mill multiple boards to the same thickness. This type of tool is critical for millwork cabinets and furniture work.

Get More Customers

Written on October 25, 2010 by Website Design Phoenix

Filed Under: Business, Marketing, Online, Small business

If you are looking to get more customers, then you need to get you business in front of the customers when they need your product or service.

Many companies spend thousands of dollars every month for TV ads, Radio spots, mailers and other forms of advertising so that their names are floating around as often as possible in the hopes that when a potential customer needs something, they will find their business.

It makes much better sense, and cents too, if you have a targeted audience that is looking for your service and are ready to buy.

But how can you do this?

Get a good website design on the first page of the search engine search results! More than fifty percent of people that are looking for a product or service go directly to Google, Yahoo!, Bing, or any of the other search engines. So if 50% of customers are looking on the internet- don’t you think you should be prominently displayed?

You can, with some specific efforts, get in the organic (non-paid) search results, you can pay for ads, and get registered (and ranked) in places like Google Maps (Google Places). Website design is critical to do this, as is having a search engine specialist for local business online advertising.

Go get your company visible online and bring in more customers for less advertising budget!

American Kennel Club - Miniature Schnauzer

Miniature Schnauzer Breed Standard

Terrier Group

General Appearance
The Miniature Schnauzer is a robust, active dog of terrier type, resembling his larger cousin, the Standard Schnauzer, in general appearance, and of an alert, active disposition. Faults - Type - Toyishness, ranginess or coarseness.

Size, Proportion, Substance
Size - From 12 to 14 inches. He is sturdily built, nearly square in proportion of body length to height with plenty of bone, and without any suggestion of toyishness. Disqualifications - Dogs or bitches under 12 inches or over 14 inches.

Head
Eyes - Small, dark brown and deep-set. They are oval in appearance and keen in expression. Faults - Eyes light and/or large and prominent in appearance. Ears - When cropped, the ears are identical in shape and length, with pointed tips. They are in balance with the head and not exaggerated in length. They are set high on the skull and carried perpendicularly at the inner edges, with as little bell as possible along the outer edges. When uncropped, the ears are small and V-shaped, folding close to the skull.

Head strong and rectangular, its width diminishing slightly from ears to eyes, and again to the tip of the nose. The forehead is unwrinkled. The topskull is flat and fairly long. The foreface is parallel to the topskull, with a slight stop, and it is at least as long as the topskull. The muzzle is strong in proportion to the skull; it ends in a moderately blunt manner, with thick whiskers which accentuate the rectangular shape of the head. Faults - Head coarse and cheeky. The teeth meet in a scissors bite. That is, the upper front teeth overlap the lower front teeth in such a manner that the inner surface of the upper incisors barely touches the outer surface of the lower incisors when the mouth is closed. Faults - Bite - Undershot or overshot jaw. Level bite.

Neck, Topline, Body
Neck - strong and well arched, blending into the shoulders, and with the skin fitting tightly at the throat. Body short and deep, with the brisket extending at least to the elbows. Ribs are well sprung and deep, extending well back to a short loin. The underbody does not present a tucked up appearance at the flank. The backline is straight; it declines slightly from the withers to the base of the tail. The withers form the highest point of the body. The overall length from chest to buttocks appears to equal the height at the withers. Faults - Chest too broad or shallow in brisket. Hollow or roach back.

Tail set high and carried erect. It is docked only long enough to be clearly visible over the backline of the body when the dog is in proper length of coat. Fault - Tail set too low.

Forequarters
Forelegs are straight and parallel when viewed from all sides. They have strong pasterns and good bone. They are separated by a fairly deep brisket which precludes a pinched front. The elbows are close, and the ribs spread gradually from the first rib so as to allow space for the elbows to move close to the body. Fault - Loose elbows.

The sloping shoulders are muscled, yet flat and clean. They are well laid back, so that from the side the tips of the shoulder blades are in a nearly vertical line above the elbow. The tips of the blades are placed closely together. They slope forward and downward at an angulation which permits the maximum forward extension of the forelegs without binding or effort. Both the shoulder blades and upper arms are long, permitting depth of chest at the brisket.

Feet short and round (cat feet) with thick, black pads. The toes are arched and compact.

Hindquarters
The hindquarters have strong-muscled, slanting thighs. They are well bent at the stifles. There is sufficient angulation so that, in stance, the hocks extend beyond the tail. The hindquarters never appear overbuilt or higher than the shoulders. The rear pasterns are short and, in stance, perpendicular to the ground and, when viewed from the rear, are parallel to each other. Faults - Sickle hocks, cow hocks, open hocks or bowed hindquarters.

Coat
Double, with hard, wiry, outer coat and close undercoat. The head, neck, ears, chest, tail, and body coat must be plucked. When in show condition, the body coat should be of sufficient length to determine texture. Close covering on neck, ears and skull. Furnishings are fairly thick but not silky. Faults - Coat too soft or too smooth and slick in appearance.

Color
The recognized colors are salt and pepper, black and silver and solid black. All colors have uniform skin pigmentation, i.e. no white or pink skin patches shall appear anywhere on the dog.

Salt and Pepper - The typical salt and pepper color of the topcoat results from the combination of black and white banded hairs and solid black and white unbanded hairs, with the banded hairs predominating. Acceptable are all shades of salt and pepper, from light to dark mixtures with tan shadings permissible in the banded or unbanded hair of the topcoat. In salt and pepper dogs, the salt and pepper mixture fades out to light gray or silver white in the eyebrows, whiskers, cheeks, under throat, inside ears, across chest, under tail, leg furnishings, and inside hind legs. It may or may not also fade out on the underbody. However, if so, the lighter underbody hair is not to rise higher on the sides of the body than the front elbows.

Black and Silver - The black and silver generally follows the same pattern as the salt and pepper. The entire salt and pepper section must be black. The black color in the topcoat of the black and silver is a true rich color with black undercoat. The stripped portion is free from any fading or brown tinge and the underbody should be dark.

Black - Black is the only solid color allowed. Ideally, the black color in the topcoat is a true rich glossy solid color with the undercoat being less intense, a soft matting shade of black. This is natural and should not be penalized in any way. The stripped portion is free from any fading or brown tinge. The scissored and clippered areas have lighter shades of black. A small white spot on the chest is permitted, as is an occasional single white hair elsewhere on the body.

Disqualifications - Color solid white or white striping, patching, or spotting on the colored areas of the dog, except for the small white spot permitted on the chest of the black.

The body coat color in salt and pepper and black and silver dogs fades out to light gray or silver white under the throat and across the chest. Between them there exists a natural body coat color. Any irregular or connecting blaze or white mark in this section is considered a white patch on the body, which is also a disqualification.

Gait
The trot is the gait at which movement is judged. When approaching, the forelegs, with elbows close to the body, move straight forward, neither too close nor too far apart. Going away, the hind legs are straight and travel in the same planes as the forelegs.

Note - It is generally accepted that when a full trot is achieved, the rear legs continue to move in the same planes as the forelegs, but a very slight inward inclination will occur. It begins at the point of the shoulder in front and at the hip joint in the rear. Viewed from the front or rear, the legs are straight from these points to the pads. The degree of inward inclination is almost imperceptible in a Miniature Schnauzer that has correct movement. It does not justify moving close, toeing in, crossing, or moving out at the elbows.

Viewed from the side, the forelegs have good reach, while the hind legs have strong drive, with good pickup of hocks. The feet turn neither inward nor outward.

Faults - Single tracking, sidegaiting, paddling in front, or hackney action. Weak rear action.

Temperament
The typical Miniature Schnauzer is alert and spirited, yet obedient to command. He is friendly, intelligent and willing to please. He should never be overaggressive or timid.

Disqualifications
Dogs or bitches under 12 inches or over 14 inches.
Color solid white or white striping, patching, or spotting on the colored areas of the dog, except for the small white spot permitted on the chest of the black. The body coat color in salt and pepper and black and silver dogs fades out to light gray or silver white under the throat and across the chest. Between them there exists a natural body coat color. Any irregular or connecting blaze or white mark in this section is considered a white patch on the body, which is also a disqualification.

MSN Careers - 4 Things That Can Increase Your Chances of a Promotion - Career Advice Article

Things That Can Increase Your Chances of a Promotion

By Robert Half International

With some companies beginning to emerge from the recession and pursue growth plans again, moving up the corporate ladder could be on your mind. Taking on a higher-level position -- and, ideally, the corresponding bump in pay -- could be especially appealing if you've assumed added responsibilities or put in longer hours during the downturn. What can you do to ensure you're on the short list of candidates for a promotion? Here are some tips:

1. Talk to your manager.
Your supervisor has a large say in your chances of earning a promotion. Don't assume he or she is aware of your desire to grow your career. Speak to your boss about your long-term plans. Your manager may know of opportunities within the department, or elsewhere in the company, and be able to help you evaluate and prepare for the opportunities. Your boss also has a more objective view of your professional strengths and weaknesses than you do and can provide an honest assessment of what steps you may need to take to assume a new role.

By involving your boss in your quest for a promotion, you can discuss with him or her any questions or challenges you face. For example, if you are an administrative assistant in the marketing department and are interested in a position as an account executive, your supervisor's feedback could help you determine if such a transition would be right for you.

2. Upgrade your skills.
There's a good chance you may lack some of the necessary skills or qualifications for a higher-level role. The supervisory position you're eyeing, for instance, may require knowledge of budgeting procedures or recruiting techniques. In some cases, you may even have to earn a new certification, license or degree.

You also need to consider if you have the necessary soft skills. A promotion may require you to do less hands-on, technical work and instead manage people, oversee projects, help various groups reach consensus or negotiate with vendors. If you lack experience in these areas, work with your manager to identify the appropriate training opportunities. Also, volunteer for projects that allow you to build skills in essential areas.

3. Expand your network. Being connected at work has multiple benefits. Most obviously, you could learn of opportunities you might not have otherwise been aware of. You also may meet someone who can provide insight into the job or type of position you seek. A manager, for example, could outline the steps he took to move up from a staff-level role and warn you about any stumbling blocks he faced.

Perhaps most importantly, networking with others allows you to boost your visibility within the company, increasing the likelihood that someone considers you when a position becomes available.

4. Gauge your interest. The most crucial part of preparing for a promotion is determining if you truly want it. While it's safe to assume that you want any additional compensation or perks the job offers, do you also want the additional responsibilities? After you've learned more about a role you're interested in assuming, ask yourself if you can do everything the position requires. Take into account not only your skill set and qualifications but also your life outside of work. If you have young children, for example, will your work/life balance suffer? Will a higher-level position require you to play office politics? Are you comfortable transitioning from peer to manager if you take a supervisory role?

A promotion is a great way to advance your career, but remember that it isn't an all-or-nothing proposition. You're unlikely to jump multiple rungs on the corporate ladder in a single move. So keep your eye on the position you want to assume eventually and focus on the steps, even if they seem small, that will allow you to get there.

Well, I took the day off and watched some old random movies on Netflix.  It is great to be able to get the old TV shows on the instant queue and turn your brain off and kind of get all nostalgic.

In fact I watch it more than I do any of my premium cable channels- so I may just get rid of them for a while- until my favorite series come back on.

Getting ready for halloween

Well, today I sat around and watched a few horror movies from the 1970s just to get in the mood for Halloween.  They were not your typical movies like Halloween, Friday 13th , or Carrie.  I watched The Last Hose on the Left from 1972 and also Burnt Offerings from 1976.  Horror was definitely a different thing back in the 70s.  It was fun to watch them today.  Maybe I will work my way through the decades for the next few days until Halloween night!

Great day out there

It has been such a beautiful day out there all day and I have been trapped inside working on the computer all day.   What a bummer!

But I did get a lot of work done and am now ready to start on a big project.  Wish me Luck!

Pool winterization. Tips on closing a swimming pool.

Fall Pool Closings

Snowbelt
  1. Balance the water chemistry:

Approximately 3 - 7 days prior to closing the pool, adjust your water balance within the ranges below:

    • pH: 7.2 - 7.6

    • Alkalinity: 80 - 120 ppm

    • Calcium Hardness: 180 - 220 ppm

Then, shock the pool with a Chlorine Shock or a Non-Chlorine Shock, at least 1lb per 10,000 gallons (follow package directions). Allow the chlorine level to return to 1.0 - 3.0 ppm before adding any winter algaecide and your pool cover. Chlorine tends to break down both algaecides and [floating] pool covers.

  2. Remove skimmer baskets, wall fittings, cleaners, solar blankets, ladders from the pool.

Put these in a safe location during the winter. Don't coil pool cleaners hoses tight.

  3. Lower water level in pool.

Using the filter pump, or a submersible pump, lower the level 12" - 18" below the skimmer for mesh covers, and 3" - 6" below the tile for solid, floating covers. If you are using an Aquador skimmer cover on aboveground or inground pools for some vinyl lined pools, the water level will not need to be lowered.

  4. Drain all pumping, filtering, heating and chlorinating equipment.

Every pump, filter, heater and chlorinator has drain plugs to allow water to drain out. All water must be drained or blown out or it will freeze and crack. After draining, D.E. filter grids or Cartridge filters should be removed and cleaned thoroughly. If the filter and pump is small enough to remove it and store it indoors, this may be desirable. If not, using a small amount of air from a shop vacuum, compressor or Mighty Vac is good to blow out any water that may still be in the equipment.

  5. Lubricate

Fall closing of the pool is a good time to lubricate the pump lid o-ring o-rings with Magic Lube. If you have a push-pull valve (also known as a slide valve) on the filter, lubricate it's o-rings as well. If you have a gas heater with cast iron plugs, lubricate these threads or leave the plugs in after draining to prevent rusting.

  6. Clean pool

Skim pool, vacuum pool, brush pool. Leaf rake (bag) types skim nets are best. Also useful for scooping large amounts of leaves/debris from pool floor. If pool is especially silty or has lots of algae, Vacuum Pool to Waste. This means to bypass the filter, and vacuum dirt from floors/walls out the backwash line. This prevents constant clogging/cleaning of filter. Place the multiport filter valve on drain to waste position (usually 2pm, if viewed as a clock face) If you have a push-pull filter valve, or a cartridge type filter there is no easy way to vacuum to waste, except for cutting the pipe coming out of the pump and then reconnecting afterwards. Brush the pool thoroughly. The pool should be as clean and clear as possible before covering.

  7. Winterize the plumbing to and from the pool

If you have an inground pool, you should blow out the lines using a Mighty Vac to blow air from the skimmer, through the equipment, and back to the pool. Then plug the lines at the pool using expansion plugs. If you don't blow the lines, add Swimming Pool Antifreeze into the line (follow package directions). Above ground pools usually just need to disconnect the hoses to and from the pump and filter, and plug the wall outlets.

  8. Add winterizing algaecide and other floaters.

Remember not to add algaecide and shock at the same time. This tends to result in the chlorine breaking down the algaecide. High chlorine levels can also be harsh to floating solid pool covers. For winter chemical kits, click here.

  9. Cover the pool.

A tight fit of your pool cover is essential. Your cover should not have holes or gaps where leaves and debris may enter the pool. A mesh safety cover provides the highest protection and safety. Solid pool covers are not safe and will require a cover pump or siphon to remove rain water and snow melt. Water Bags or AquaBloks are used to secure an inground solid pool cover. Above ground pool covers use a cable/winch device to secure the cover around the pool. Air Pillows are used in above ground pools to absorb the expansion of ice inside the pool. In areas of high wind, an above ground pool owner will find wall bags or cover seal useful products. A leaf net is very useful if you have a lot of trees surrounding your pool.

Sunbelt

In areas where it rarely gets below freezing temperatures, many people simply reduce the amount of filtration time per day, and also will find that the pool needs fewer chemicals. Covering the pool, even if not winterizing, will reduce cleaning and chemical and filtering demand even further.

New Apple MacBook Air costs less, plus App Store is coming - USATODAY.com

CUPERTINO, Calif. — Apple (AAPL), basking in the success of the iPhone and iPad, is bringing the look of those mobile devices to its computers.

Apple CEO Steve Jobs introduced a MacBook Air notebook, which — like its popular iPad — has no hard drive. It relies instead on flash memory, which is pricey but more durable if dropped or bumped. It also has a much longer battery life

Follow this link to learn more about the MacBook Air and the App Store for home computers, not just phones!!

Car Maintenance Service

Well, I have been sitting in a Borders book store all day working on the wifi while I wait for my Nissan truck to be serviced.  Had to get the 45,000 mile service done and then there was an issue with a sensor in the gas tank going bad.  Luckily Nissan knew about the issue and they extended the warranty!  Still had to pay out the nose for the service though.

Officials in a Mexico City borough said 600 residents made it into the Guinness Book of World Records by creating the world’s largest enchilada.

Guinness officials said the Iztapalapa borough’s enchilada, measuring 230 feet, 12 inches in diameter, beat earlier attempts elsewhere in the country and in the United States, La Jornada reported Monday.

The next Blue Man Group?!

I have to say that some of the more strange things that I tend to find on the internet seem to originate from Japan.

This little video is a Japanese version of a sadomasochistic Blueman group.

It is really kind of a strange thing.  At first I thought the uh... drums... were just plastic forms but then I took a look at the red boxes and realized that there was more to it.  Once the screaming started, I actually laughed my "drum" off!

Go take a look to get your groove on!
